RESUMO

The circadian clock coordinates the daily rhythmicity of biological processes, and its dysregulation is associated with various human diseases. Despite the direct targeting of rhythmic genes by many prevalent and World Health Organization (WHO) essential drugs, traditional approaches can't satisfy the need of explore multi-timepoint drug administration strategies across a wide range of drugs. Here, droplet-engineered primary liver organoids (DPLOs) are generated with rhythmic characteristics in 4 days, and developed Chronotoxici-plate as an in vitro high-throughput automated rhythmic tool for chronotherapy assessment within 7 days. Cryptochrome 1 (Cry1) is identified as a rhythmic marker in DPLOs, providing insights for rapid assessment of organoid rhythmicity. Using oxaliplatin as a representative drug, time-dependent variations are demonstrated in toxicity on the Chronotoxici-plate, highlighting the importance of considering time-dependent effects. Additionally, the role of chronobiology is underscored in primary organoid modeling. This study may provide tools for both precision chronotherapy and chronotoxicity in drug development by optimizing administration timing.

Most Alzheimer's disease drugs do not work efficiently because of the blood-brain barrier. Therefore, we designed a new nanopreparation (PS-DZP-CHP): cholesterol-modified pullulan (CHP) nanoparticle with polysorbate 80(PS) surface coverage, as donepezil (DZP) carrier to realize brain tissue delivery. By size analysis and isothermal titration calorimetry, we chose the optimal dosing ratio of the drug with nanomaterials (1:5) and designed a series of experiments to verify the efficacy of the nanoparticles. The results of in vitro release experiments showed that the nanoparticles can achieve continuous drug release within 72 h. The results of fluorescence observation in mice showed a good brain targeting of PS-DZP-CHP nanoparticles. Furthermore, the nanoparticle can enhance the drug in the brain tissue concentration in mice. DZP-CHP nanoparticles were used to pretreat nerve cells with Aß protein damage. The concentration of lactate dehydrogenase was determined by MTT, rhodamine 123 and AO-EB staining, which proved that DZP-CHP nanoparticles had a protective effect on the neurotoxicity induced by Aß25-35 and were superior to free donepezil. Microthermal perpetual motion meter test showed that PS-DZP-CHP nanoparticles have an affinity with apolipoprotein E, which may be vital for this nanoparticle targeting to brain tissue.

For lithium-sulfur batteries, commercial application is hindered by the insulating nature of sulfur and the dissolution of the reaction intermediates of polysulfides. Here, we present an ordered meso-microporous core-shell carbon (MMCS) as a sulfur container, which combines the advantages of both mesoporous and microporous carbon. With large pore volume and highly ordered porous structure, the "core" promises a sufficient sulfur loading and a high utilization of the active material, while the "shell" containing microporous carbon and smaller sulfur acts as a physical barrier and stabilizes the cycle capability of the entire S/C composite. Such a S/MMCS composite exhibits a capacity as high as 837 mAh g(-1) at 0.5 C after 200 cycles with a capacity retention of 80% vs the second cycle (a decay of only 0.1% per cycle), demonstrating that the diffusion of the polysulfides into the bulk electrolyte can be greatly reduced. We believe that the tailored highly ordered meso-microporous core-shell structured carbon can also be applicable for designing some other electrode materials for energy storage.

A lithium-oxygen battery would deliver the highest energy density of a rechargeable battery, but the multiphase electrochemical reaction on the air cathode has difficulty proceeding when operated with only solid catalysts. We report here the organic-electrolyte-dissolved iron phthalocyanine (FePc) as a shuttle of (O2)(-) species and electrons between the surface of the electronic conductor and the insulator Li2O2 product of discharge. The Li2O2 is observed to grow and decompose without direct contact with carbon, which greatly enhances the electrochemical performance. Our results signal that the use of molecular shuttles that are catalytically active may prove to be enablers of a practical lithium-air rechargeable battery.

Lithium-sulfur batteries have great potential for some high energy applications such as in electric vehicles and smart grids due to their high capacity, natural abundance, low cost and environmental friendliness. But they suffer from rapid capacity decay and poor rate capability. The problems are mainly related to the dissolution of the intermediate polysulfides in the electrolyte, and to the poor conductivity of sulfur and the discharge products. In this work, we propose a novel dual coaxial nanocable sulfur composite fabricated with multi-walled nanotubes (MWCNT), nitrogen-doped porous carbon (NPC) and polyethylene glycol (PEG), i.e. MWCNTs@S/NPC@PEG nanocable, as a cathode material for Li-S batteries. In such a coaxial structure, the middle N-doped carbon with hierarchical porous structure provides a nanosized capsule to contain and hold the sulfur particles; the inner MWCNTs and the outer PEG layer can further ensure the fast electronic transport and prevent the dissolution of the polysulfides into the electrolyte, respectively. The as-designed MWCNT@S/NPC@PEG composite shows good cycling stability and excellent rate capability. The capacity is retained at 527 mA h g(-1) at 1 C after 100 cycles, and 791 mA h g(-1) at 0.5 C and 551 mA h g(-1) at 2 C after 50 cycles. Especially, the high-rate capability is outstanding with 400 mA h g(-1) at 5 C.
